Effects of Carthami Flos, Moutan Radicis Cortex, Scirpi Rhisoma on Cellular Activity of Gingival Fibroblasts and Periodontal Ligament Cells


Abstract
Peirodontal disease was chronic imflammatory disease and was appeared when pathogens suppressed the host difense mechnism. Clinical signs were characterized as pain, pus discharge, tooth mobility, peridontal pocket formation.
A principle etiology was gram-negative bacteria. To eliminate the etiologic factors, non-surgical and surgical procedure were used. Adjuvant drugs were effecacious to favorable process of healing and anti-inflammation.
Many antibiotics were used, today, side effect were appeared such as host tolerance to drugs and antibiotics-risistance bacteria. Therefore, new drugs are needed. Carthami Flos, Moutan Radicis Cortex, Scirpi Rhisoma is used in oriental medicine for anti-inflammatory, vascular circulation, analgesic and sedative effect.
In present study, we examined the effect of Carthami Flos, Moutan Radicis Cortex, Scirpi Rhisoma on Cellular Activity of gingival fibroblasts and periodontal ligament cells.
To evaluate the effect of Carthami Flos, Moutan Radicis Cortex, Scirpi Rhisoma on Cellular Activity of gingival fibroblasts, the cells were seeded at a cell density of 2 x 103 cells/well in 96-well culture plates. After one day incubation, 10-¢¥, 102, 10-3 and 10¢¥g/mf of Carthami Flos, Moutan Radicis Cortex, Scirpi Rhisoma were added to the each and incubated for 1, 2 and 3 days, respectively. The MTT assay were carried out.
To evaluate the effect of Carthami Flos, Moutan Radicis Cortex, Scirpi Rhisoma on Cellular Activity of periodontal ligament cells, the cells were seeded at a cell density of 2 x 103 cells/well in 96-well culture plates. After one day incubation, 10-2, 10-3, 104 and 10-Sg/mi of Carthami Flos, Moutan Radicis Cortex, Scirpi Rhisoma were added to the each and incubated for 1, 2 and 3 days, respectively. The MTT assay were carried out.
The results were as follows
1. Carthami Flos, Moutan Radicis Cortex, Scirpi Rhisoma were significantly increased cellular activity of gingival fibroblasts and periodontal ligament cells than control at specific concentration and day in main experiment.
2. Generally, Carthami Flos, Moutan Radicis Cortex, Scirpi Rhisoma were high cellular activity of gingival fibroblasts than periodontal ligament cells at same concentraton and day in main experiment.
These findings suggest that Carthami Flos, Moutan Radicis Cortex, Scirpi Rhisoma increase cellular activity at specific concentration(10-3g/Mi). Carthami Flos, Moutan Radicis Cortex, Scirpi Rhisoma are respected to help healing process after periodontal treatment. Clinical application of Carthami Flos, Mot-an Radicis Cortex, Scirpi Rhisoma may need to further studies.
